원리

  

  • 3-4 개의 siRNA로 이루어진 pool은 종종 multiple off-target gene에 영향을 주는 반면, 30개의 siRNA Mixture로 이루어진 siPOOL은 picomolar에서 작용하며, on-target specificity를 높여줍니다.

 

 

 

 

실험 결과

 

 

   

Off-target effect를 낮출 수 있습니다.  

(Transcriptome-wide profiling에 의하면 single siRNA가 여러 off-target genes(red dots)를 유발하는 결과가 나옴. 반면에 같은 target(green dot)에 대해 siPOOL이 off-target effect를 감소시켰음.)


 

 

 

 

일관적인 target Knock-down 효과를 보여줍니다. 

(개별 single siRNA는 knock-down 효율의 편차가 큼. 같은 target에 대해 두 개의 다른 siPOOLs가 비슷한 knockdown efficiencies을 보임.)

 

 

 


 

siPOOLs로 일관적인 phenotypes를 재현할 수 있습니다.

(36 genes screening하였는데 gene당 두개의 siPOOL 과 commercial 3개의 siRNAs로 cell viability에 대한 phenotype을 분석하였음. siPOOLs가 더 일관적인 phenotype을 보여주었음.)
