  • Tocris에서는 ISO TS 20399 지침을 따라 세포치료제 제조시 보조물질로 이용되는 small molecule을 보조물질 등급 (Ancillary Material, AM)으로 제공합니다.
  • 보조 물질은 최종 제품에서는 발견되지 않지만 치료제가 clinical use로 이용될 수 있도록 제품의 적합성에 영향을 미칠 수 있으므로 등급이 중요합니다.

A 83-01 blocks phosphorylation of Smad2 and inhibits TGF-β-induced epithelial-to-mesenchymal transition. Only weakly inhibits ALK-1, -2, -3, -6 and MAPK activity. More potent than SB 431542 (Cat. No. 1614). A 83-01 inhibits differentiation of rat induced pluripotent stem cells (riPSCs) and increases clonal expansion efficiency. Helps maintain homogeneity and long-term in vitro self-renewal of human iPSCs. Also promotes neural differentiation of hPSCs as part of a chemical cocktail.
TB5758-RMU AGN 193109  AGN 193109 is a high affinity pan-retinoic acid receptor (RAR) antagonist (Kd values are 2, 2 and 3 nM at RARα, RARβ and RARγ, respectively). Exhibits no significant affinity for retinoic X receptors. AGN 193109 promotes the differentiation from mouse embryonic stem cells into paraxial mesoderm. AGN 193109 suppresses primary human cardiovascular smooth muscle cell calcification.
TB0760-RMU AM 580  AM 580 is an analog of retinoic acid that acts as a selective RARα agonist (EC50 values are 0.3, 8.6 and 13 nM for RARα, RARβ and RARγ respectively). Significantly induces IL-4, IL-5 and IL-13 and inhibits IL-12 and IFNγ synthesis, and induces cell differentiation with over 7 times the activity of retinoic acid in vitro.
TB2634-RMU DAPT DAPT is a γ-secretase inhibitor. DAPT reduces Aβ40 and Aβ42 levels in human primary neuronal cultures (IC50 values are 115 and 200 nM for total Aβ and Aβ42 respectively) and in brain extract, cerebrospinal fluid and plasma in vivo. DAPT has no effect on APPα and APPβ levels. DAPT blocks Notch signaling in hybrid human-mouse fetal thymus organ culture (FTOC) and causes ESCs to commit to neuronal differentiation. TB4489-RMU DBZ  DBZ is an inhibitor of γ-secretase aspartyl protease. DBZ blocks amyloid precursor protein-like (APPL) and Notch cleavage (IC50 values are 2.6 nM and 1.7-2.9 nM, respectively). By inhibiting Notch and the Notch pathway, DBZ improves efficiency of iPSC generation and production of iPSCs from mouse and human keratinocytes without KLF4 and CMYC. In an animal model of Alzheimer's disease, DBZ inhibits the cleavage of amyloid precursor protein into Aβ40. DBZ promotes cochlear supporting cell proliferation and hair cell mitotic regeneration in neonatal mice. DBZ in combination with iDOT1L enables maintenance of human naïve-state pluripotency in non-hypoxic conditions.
TB4703-RMU DZNep HCl  3-Deazaneplanocin A hydrochloride is a histone methyltransferase inhibitor; decreases global histone methylation. 3-Deazaneplanocin A hydrochloride inhibits EZH2 histone methyltransferase and s-adenosylhomocysteine (SAH) hydrolase activity. Blocks trimethylation of lysine 27 on histone H3 and lysine 20 on histone H4 in vitro. 3-Deazaneplanocin A hydrochloride induces apoptosis in multiple cancer cell lines and has no apoptotic effect on normal cells. TB3742-RMU RepSox RepSox is a potent and selective inhibitor of the TGF-β type I receptor/ALK5 (IC50 values are 4 and 23 nM for TGF-β type I receptor autophosphorylation and binding respectively). Selective for TGF-β type I receptor over a range of kinases, including p38 MAPK, JNK1 and GSK3 (IC50 > 16 μM). Enhances the efficiency of cellular reprogramming; replaces Sox2 by inducing Nanog expression.
TB6666-RMU T3 T3 is a thyroid hormone. Promotes differentiation of oligodendroglial precursor cells in neonatal rats and promotes maturation of hiPSC-derived cardiomyocytes. Also specifies cone cell subtype in retinal cell differentiation protocols. T3 is used in protocols to generate cortical spheroids from hPSCs.
TB4855-RMU WIKI4 WIKI4 is an inhibitor of Wnt/β-catenin signaling (EC50 ~ 75 nM). Inhibits auto-ADP-ribosylation of tankyrase 2 (TNKS2) (IC50 ~15 nM); prevents the ubiquitination and degradation of axin.
