줄기세포 생존력 향상을 위한 CEPT Cocktail Kit | 고마바이오텍

CEPT란?

기초 연구와 치료 응용 분야에서 줄기 세포를 성공적으로 사용하는 데 있어 오랜 과제는 스트레스에 대한 세포의 민감성으로 인해 광범위한 세포 사멸이 발생한다는 것입니다.

CEPT Cocktail은 PSC의 생존력을 향상시키는 Chroman 1 (Cat. No. 7163), Emricasan (Cat. No. 7310), Lyophilized Polyamine Supplement x1000 (Cat. No. 7739) 및 Trans-ISRIB (Cat. No. 5284) 소분자의 조합으로, 줄기세포 실험의 모든 단계에서 다능성 줄기세포(PSC)의 생존율을 높이고, 배아체/오가노이드 형성 시기에 세포 생존률을 크게 향상시킵니다.  

CEPT Cocktail은 줄기세포 계대 과정에서 세포 스트레스와 DNA 손상을 줄이고 세포 접착을 촉진함으로써 줄기세포 보호 기능을 제공하여, 질병 모델링, 약물 스크리닝, 재생 의학 및 세포 치료를 위한 세포 제조를 개선할 수 있는 광범위한 잠재력을 가지고 있습니다. 

특징 

• Chroman 1 (#7163)은 NCATS 팀에 의해 potential & selective ROCK inhibitor로 발견되었으며, 단독으로, 혹은 다른 small molecule과 함께 사용했을 때  세포의 생존률을 크게 향상시킵니다.
• Emricasan (#7310)은 Chroman 1과 함께 PSC의 apoptosis를 줄여줍니다.
• Trans-ISRIB (#5284)은 단백질 합성 및 세포 passaging시 cell attachment를 돕습니다.
• Polyamine (#7739)은 cell growth에 필수적이며, cellular structure와 macromolecules를 안정화시키고, transcription, translation, cell cycle, stress response등과 같은 기능을 중재합니다. 
• PSC culture시 polyamine이 Chroman 1, Emricasan, Trans-ISRIB과 함께 사용될 경우, polyamine은 cell attachment와 protein translation을 크게 향상시켜줍니다. 
• CEPT는 long-term passaging, single cell cloning, gene editing, PSC differentiation시 cell survival을 향상시켜줍니다. 
• 또한 differentiated cell의 냉동 보관 후에도 세포 생존력을 증가시켜줍니다. DMSO Control과 비교했을 때, Cardiomyocyte의 생존률은 36%, motor neuron 생존율은 63%까지도 증가하였습니다. 
• large-scale의 hiPSC 생산 및 분화세포 표준화에도 매우 효과적입니다.
