아데노바이러스는 선형 이중 가닥 DNA를 가진 non-toxic, non-integrating, non-enveloped 바이러스입니다. 아데노바이러스 벡터는 Ad5 혈청형을 기반으로 합니다. 세포에 들어가기 위해 Ad5 기반 벡터는 Coxsackie-Adenovirus Receptor(CAR)를 사용합니다. 재조합 벡터의 패키징 용량을 늘리기 위해 E1 및 E3 유전자가 삭제됩니다. E1은 패키징 세포(HEK293)에 의해 제공되며 E3는 바이러스 생산에 필수적이지 않습니다. 바이러스 벡터에서 E1을 삭제하면 바이러스 복제가 방지됩니다.

아데노바이러스 벡터의 생산은 BAC 시스템을 사용한 rAdV 플라스미드 구성을 기반으로 합니다. 관심 유전자(GOI)/shRNA의 cDNA는 먼저 과발현(OEX) 또는 녹다운(KD)을 위해 AV 셔틀 플라스미드에 클로닝된 다음 pAD5 벡터 유전자를 포함하는 BAC 벡터를 포함하는 BA5-FRT 세포로 변환됩니다. FLP 매개 BAC 벡터 재조합이 진행됩니다. Pac 1 digestion은 BAC-DNA를 제거하고 선형 Ad5-GOI 구조를 생성합니다.

이 선형 구조물은 HEK 292 또는 CAP producer cell로의 형질감염에 의해 GOI를 발현하는 재조합 아데노바이러스 벡터로 재구성됩니다. 이렇게 하면 기본 저역가 바이러스 stock이 생성됩니다. 바이러스 역가는 필요한 규모로 증폭되고 농축되며 QC 테스트를 위해 정제됩니다. 그런 다음 GOI가 포함된 아데노바이러스 입자가 채워지고 배송 준비가 완료됩니다.

The cells are incubated for 2-3 days, then virus is collected from the supernatant and cell lysates. The virus collected from the first round is used for the infection of the cells in the 2nd round and for the larger amount of virus the 3rd round is used. The Ad-GOI is further purified and concentrated by CsCl (large-scale production) or using AdenoOne purification kit (small-scale productions).
