구성

• Annexin V-FITC
• 5X Binding buffer
• Propidium Iodide (PI)

실험 방법

1.  분석하고자 하는 세포를 약1x10^6cells/ml로 준비한다.
2.  0.5ml의 세포 현탁(5x10^5cell/ml)을 microtube로 옮긴다.
3.  1000 x g로 5분간 상온에서 원심 분리하여 배양액을 제거한다.
4.  차가운 PBS 0.5ml로 세포를 부드럽게 풀어준다.
5.  1000 x g로 5분간 상온에서 원심 분리하여 PBS를 제거한다.
6.  차가운 1x binding buffer 0.5ml로 세포를 부드럽게 풀어준다.
7.  1.25ul의 annexin V-FITC를 가한다.
8.  반응물에 빛을 차단하고 상온에서 15분 반응시킨다.
9.  1000 x g로 5분간 상온에서 원심 분리하여 상층액을 제거한다.
10.   차가운 1x binding buffer 0.5ml로 세포를 부드럽게 풀어준다.
11.   10ul의 propidium iodide를가한다.
12.   반응물을 빛을 차단하고 냉장 보관한다 (4℃)
13.   빠른 시간 내에 유세포분석 (flow cytometry)또는 형광현미경으로 분석 한다.
