특징

 

  • 박테리아는 His나 GST tag이 붙은 재조합 단백질 발현에 널리 사용됩니다. soluble form의 GST fusion protein은 크로마토그래피에 의해 박테리아 용해물로부터 정제되는데, 재조합 단백질이 inclusion body를 형성했을 경우, Rapid GST Inclusion Body Solubilization and Renaturation Kit을 이용하여 가용성 형태 (soluble form)으로 복원시켜 줍니다.
  • GST inclusion body를 2시간 이내에 가용화 및 복원시킵니다.
  • 장시간의 투석 혹은 희석 단계가 필요 없습니다.
  • pH의 변화 또는 산화-환원에 영향을 받지 않아 사용이 간편합니다.
  • 박테리아 배양액 5-10 리터를 가용화 및 복원시킬 수 있는 충분한 양의 시약들이 포함되어 있습니다.
  • 기존의 GuHCl 이나 Urea solubilization 과 refolding method에 비해 시간이 절약되고 pH variation이 없습니다.

 Solubilization_and_Renaturation_of_GST-RTK_fusion_protein 

 

Solubilization and Renaturation of GST-RTK fusion protein. GST-RTK expression was induced with 1 mM IPTG at 37ºC for 4 hours. The cell pellet was lysed, and inclusion bodies were solubilized and renatured under different amounts of detergent solubilization solution according to the assay protocol. Lane 1: MW Standard; Lane 2: Whole E. coli lysate; Lanes 3, 7, 11: No detergent; Lanes 4, 8, 12: 32-fold dilution; Lanes 5, 9, 13: 8-fold dilution; Lanes 6, 10, 14: 2-fold dilution.
