특징

Molecular Glue는 Ubiquitin E3 ligase로 표적 단백질의 이량체화 (dimerization)을 유도하여 표적 단백질 분해를 달성하는 소분자입니다.
이량체화에 의한 분해 효과는 E3 ligase와 관심 단백질의 직접적인 결합 상호작용을 통해 일어날 수도 있고 E3 ligase 단백질 구조의 allosteric 변형에 의해 일어날 수도 있습니다.
E3 ligase, 관심 단백질 및 소분자로 구성된 complex의 형성은 관심 단백질의 유비퀴틴화를 초래하고 프로테아좀에 의한 파괴를 촉발합니다. 유비퀴틴화 단계 후에 분자 접착제가 해리되어 새로운 단백질 복합체를 형성하게 됩니다.
Molecular Glue는 타겟 단백질을 분해하기 위해 PROTAC 분자와 유사한 방식으로 작동하지만, PROTAC degrader와달리 chimeric molecule이 아닙니다.
