특징

• 미토콘드리아 기능 장애는 급성 췌장염, 허혈-재관류 손상, 근이영양증, 신경변성 등의 발병을 초래합니다. 
• 원인은 미토콘드리아 투과성 전이 구멍 (mitochondrial permeability transition pore (mPTP))이 지속적으로 개방되어 내부 미토콘드리아 막의 투과성이 증가하여 ATP 생산이 중단됨으로써 결국 세포사멸을 초래하게 됩니다.
• mPTP의 opening과 inhibition은 미토콘드리아의 calcium retention capacity (CRC)를 측정하는 Calcium Green-5N을 이용하여 평가합니다. 
  
• 5μM 염화칼슘을 5분 간격으로 digitonin 투과성 세포에 10회 첨가하고, 각각 매트릭스에서 칼슘 흡수로 인해 소멸되는 신호의 스파이크를 유도합니다. mPTP가 열리면 미토콘드리아 막 전위가 붕괴되어 매트릭스에서 축적된 칼슘이 방출되어 형광 신호가 갑자기 지속적으로 증가합니다.
  
• CRC 분석은 잠재적인 mitochondrial pore inhibitor나 inducer는 HepG2 (human hepatocytoma) 또는 SH-SY5Y (human neuroblastoma) cell을 이용하여 FLIPR TETRA를 이용하여 96-well 형식으로 실행됩니다.
