특징

 

  • 혈관내피와 백혈구 또는 종양 세포의 상호 작용은 세포가 내피세포의 접착분자 (endothelial cell adhesion molecule)에 부착하는 일련의 과정으로 구성됩니다.
  • CytoSelect™ Leukocyte Endothelium Adhesion Assay는 백혈구와 내피 사이의 상호작용을 정량적으로 측정하기 위한 강력한 시스템을 제공합니다.
  • 부착 세포는 형광 플레이트 판독기에서 쉽게 정량화할 수 있습니다.
  • 내피 세포들을 activator나 inhibitor로 처리후 배양하고, leukocyte를 LeukoTracker로 라벨링하여 cell에 처리하면 내피세포를 투과한 leukocyte의 수를 정량할 수 있습니다.






원리

  

  • 내피세포를 배양하여 monolayer를 형성하고, 원하는 activator나 inhibitor로 처리합니다.  백혈구는 LeukoTracker™로 표지된 후 pellet, wash, resuspend 됩니다. 백혈구를 내피세포 monolayer에 추가하고 배양한 후 부착된 백혈구를 세척, 용해 후 fluorometer로 정량합니다.

실험 결과


Quantitation of Human Monocytic THP-1. LeukoTracker™ labeled THP-1 cells were titrated in 1X PBS, then subsequently lysed with 2X Lysis Buffer  (75 µL of cell suspension was mixed with 75 µL of 2X Lysis Buffer). Fluorescence was quantified as described in Assay Protocol.

 
Human Monocytic THP-1 Adhesion to HUVEC Monolayer. HUVEC monolayer in 48-well plate was treated with 1 µM PMA for 12 hrs. LeukoTracker™ labeled THP-1 cells were allowed to attach to HUVEC monolayer for 1 hr. Adherent cells were lysed and quantified by as described in the Assay Protocol.




Cytokine Activation of Human Monocytic THP-1 Adhesion to HUVEC. HUVEC monolayer (left) or THP-1 cells (right) were treated with 50 ng/mL TNFα  or 1 µM PMA for 12 hrs. LeukoTracker™ labeled THP-1 cells (50,000 cells/well) were allowed to attach to HUVEC monolayer for 1 hr. Adherent cells were lysed and quantified by as described in the Assay Protocol.
