오토파지 연구 | Autophagy Research Small Molecules

Autophagy는 세포내 재활용 시스템이라고도 불리우며, 불필요하거나 기능하지 않는 세포 구성성분을 자연적으로 분해하는 파괴 기전입니다. Autophagy의 활성화와 저해에 관여하는 activator & inhibitor를 소개합니다.

Autophagy란?

Autophagy (오토파지, 자가소화작용)은 기아 또는 스트레스 신호에 대한 응답으로 세포가 거대 분자를 분해하는 과정입니다. 손상된 미토콘드리아와 Ubiquitin-Proteasome 시스템에 의해 분해 되기에는 너무 큰 단백질을 제거하는데 도움이 되고, 비가역적으로 손상된 세포를 제거하여 전신 항상성을 유지하는데 도움이 됩니다. 이러한 능력은 세포 생존과 신경변성 완화에 관여됩니다. autophagy는 apoptosis와 밀접하게 연관되어 있지만, 주로 세포 에너지 항상성이 유지되고 세포 소기관과 단백질이 분해되는 이화 작용 메커니즘이 특징입니다.

Autophagy는 또한 apoptosis를 유도하는 것과 유사한 스트레스에 대한 반응으로 발생하며 여러 proapoptotic 신호도 autophagy를 유도합니다. Apoptosis는 phagocytic cell lysosomes을 사용하여 사멸을 유도하고 Autophagy는 세포의 통합 리소좀 장치를 사용하여 사멸을 유도하며, 두 메커니즘의 차이점은 현미경으로 관찰할 수 있습니다.

Autophagy에 손상이 있을 경우, tau, a-synuclein, huntington 단백질 단편의 응집을 초래할 수 있으며, 신경퇴행성 질환을 야기할 수 있습니다. 또한 대사장애 (당뇨병, 비만) 및 노화에도 관련이 있으며, 암 발병에도 관여합니다.

세포내 Autophagy 조절은 mTOR와 PI 3 Kinase signaling에 의해 제어됩니다. mTORC1은 조절을 직접적으로 담당하고, Class I PI 3-K-Akt pathway에서는 autophagy 억제인자로 작용합니다. AMPK 활성화는 mTOR 의존적인 신호 전달을 억제하여 Autophagy를 자극합니다.
