• β-Arrestin은 GPCR의 활성화, 7-TM receptor의 desensitization, Protein kinase와 같은 신호 분자의 조절에 관여합니다. GPCR의 경우 ligand에 의해 유도된 β-Arrestin 신호는 G-protein signaling과 독립적인 신호 cascade를 활성화하여 antagonist mode screening, ligand pharmacology 연구, GPCR deorphanizing시 이상적인 신호를 제공합니다.

• GPCR-PK의 활성화는 2개의 β-galactosidase 효소 단편(EA 및 PK)의 complementation을 유도하고, 이렇게 생성된 기능적 효소인 β-galactosidase는 기질을 가수분해하여 화학발광 신호를 생성합니다.

• PathHunter β-Arrestin GPCR cell line은 ProLink (PK)가 tagging된 GPCR과 Enzyme Acceptor (EA)가 tagging된 β-arrestin을 함께 발현하도록 엔지니어링 되었습니다.
  
• GPCR-PK가 활성화 되면 β-Arrestin-EA가 붙으면서 EA와 PK 두개의 β-galactosidase enzyme freagment가 결합되고, 이렇게 생성된 fuctional enzyme은 substrate를 가수분해하여 chemiluminescent signal을 생성합니다.
