특징

 

  • FastGro는 FBS를 대체하는 합성 supplement로, 세포 배양 배지에 첨가하여 사용합니다.
  • 동물 유래 성분이 포함되어 있지 않으므로, serum을 사용할 때 발생 할 수 있는 unknown 단백질들로 인한 간섭현상이 없습니다.
  • Lot 마다 variation 이 없어 매 Lot 별 testing 없이도 일정한 결과를 얻을 수 있습니다.

사용 방법

1. FBS 사용할 때와 동일한 양을 배지에 넣습니다.
2. FBS를 함유하고 있는 media에서 FastGro 함유하는 media로 이전하기 위해서는 세포에 스트레스가 최소화 되도록 농도를 순차적으로 조절하며 배양해가면서 media를 교체해 줍니다.
  • FBS를 대체할 때 아래의 사항을 고려해 주세요.
        1) 혈청이 없는 조건에서는 별도의 plate coating이 필요 합니다. (recombinant coating material 필요)
        2) 혈청이 없는 경우 Trypsin을 사용하시면 세포에 damage를 일으킬 수 있습니다. (Accutase™ 등의 non-trypsin enzyme 사용을 권장 드립니다.)
        3) 혈청이 없는 경우 기존농도로 penicillin-Streptomycin을 사용하시면 세포에 과다 작용하여 damage를 일으킬 수 있습니다. (무항생제 혹은 Gentamicin 사용을 권장 드립니다.)

Figure 1. Growth evaluation in FBS-supplemented versus FastGro™-supplemented medium
This study was performed with Vero cells adapted over long-term culture in serum-free, FastGro™-supplemented medium. Growth dynamics were compared to those of the sub-line cultured in FBS-supplemented medium. The results are from a 6-day culture period in T25 flasks. Cell numbers were determined at the end of the 144-hour culture period (Figure 1).

In this study, a Vero strain was adapted over 3 passages to serum-free growth in FastGro™-supplemented Williams Medium E grown on microcarriers (200 mg cytodex/100 mL corresponding to 1200 cm2  growth area) in small-scale laboratory bioreactors. To test for potential positive effects of insulin, the hormone was added to the culture medium (1.25 mg/L) of one group from day 0 through day 4, then switched back to Williams E + FastGro™ alone.
As can be observed, in both conditions the Vero cells showed a constant growth over the entire experimental period (Figure 2). The addition of insulin during the initial growth phase (4 days) caused a growth enhancement during the logarithmic growth phase, albeit the two experimental groups showed only a slight difference (< 10%) in the final cell numbers (< 10%). These results imply that a short mitogenic stimulus by insulin during the initial growth phase may be sufficient for generating a sustained growth benefit over the whole batch culture period.
