특징

 

Comet Assay(코멧 어세이)는 단일 세포 젤 전기영동(Single Cell Gel Electrophoresis)이라고도 불립니다. 세포를 젤(Agarose)에 굳힌 뒤 전기영동을 걸면, 손상된 DNA 조각들이 핵 밖으로 빠져나와 이동합니다. 이 모양이 마치 밤하늘의 혜성(Comet) 같다고 해서 '코멧 어세이'라는 이름이 붙었습니다. 혜성의 꼬리(Tail)가 길고 밝을수록 DNA 손상이 심하다는 뜻입니다. 

 

  • 손상의 원인(방사선, 화학 물질, 자외선 등)에 상관없이 세포 내 전체적인 DNA 손상 정도를 확인할 수 있는 빠르고 편리한 스크리닝 도구입니다.
  • 단일 세포 수준에서 아주 미세한 DNA 단편화(Fragmentation)까지 감지할 수 있습니다
  • 전류가 흐르는 동안 손상된 DNA는 "comet (혜성)" 모양을 형성하며, Epifluorescence microscopy로 확인한 결과 건강한 DNA와 시각적으로 구분됩니다.
  • 3, 10, 24, 48, 96-Well의 다양한 형태로 제공됩니다. 
  • 3 / 10-Well: 소수의 샘플을 정밀하게 관찰하거나 개별 세포의 이미지를 크게 얻고 싶을 때 적합합니다.
  • 24 / 48 / 96-Well: 다수의 약물 후보군을 동시에 테스트하는 고처리량 스크리닝(High-throughput screening)에 최적화되어 있습니다.
  • 슬라이드 표면이 아가로스(Agarose) 젤과 강력하게 밀착되도록 처리되어 있으므로, 전기영동이나 세척 과정 중에 샘플 젤이 슬라이드에서 떨어져 나가는 문제가 없습니다.
  • 키트 전체뿐만 아니라 슬라이드만 따로 구매할 수 있어, 자체 시약을 가진 실험실에서도 경제적으로 사용할 수 있습니다.


적용 분야  

 

  • 독성학: 특정 약물이나 화학물질이 세포 DNA에 미치는 영향 연구.
  • 환경 연구: 오염 물질에 노출된 생물체의 유전적 손상 측정.
  • 암 연구: 항암제 처리 후 암세포의 DNA 파괴 정도 확인.

  

 

실험 원리 및 순서 

 

  1. 세포 현탁: 검사할 세포를 아가로스와 섞어 특수 슬라이드에 올립니다.
  2. Lysis (세포 용해): 세포막을 터뜨려 DNA만 남깁니다.
  3. 전기영동: 일정한 전압을 가하면 손상된(잘린) DNA는 플러스(+) 극 쪽으로 이동하며 꼬리를 형성합니다.
  4. 염색 및 관찰: DNA 전용 형광 염색약으로 염색한 뒤 형광 현미경으로 관찰합니다.
