특징  

 

• Cyclin dependent kinase(CDK)는 세포주기, 전사, 신경 기능 및 발달을 통한 진행을 제어하는 serine/threonine kinase입니다. CDK의 활성은 cyclin에 의존합니다. CDK는 사람에서 20종류가 알려져 있으며, 진화적/기능적 관계에 따라 그룹을 나누며, 29개의 cyclin에 의해 조절됩니다. 
• 세포 주기는 CDK1, CDK4와 CDK5 subfamily에 의해 주로 제어되며, 다른 CDK는 전사에서 역할을 하고 세포 주기 의존적인 방식으로 조절되지는 않습니다.

CDK1 Subfamily (CDK1, CDK2 & CDK3) Inhibitors & Degraders   

CDK1 subfamily는 CDK1, CDK2, CDK3를 포함하며 모두 세포 주기의 조절과 진행에 관여합니다. CDK1은 cyclin A나 cyclin B에 결합하고 세포 주기 중 S와 G2 phase에 중요하고, CDK2는 cyclin E에 결합하고 세포 주기 중 G1/S phase 동안의 checkpoint control protein으로 작용합니다. CDK1이나 CDK2의 억제는 세포 주기를 중단하고 apoptosis로 이어지게 합니다.

CDK4 Subfamily (CDK4 & CDK6) Inhibitors & Degraders 

CDK4와 CDK6은 세포 주기 중 G1 phase를 조절하는 사이클린 의존적 인산화효소와 밀접하게 관련되어 있습니다. CDK4는 모든 세포에서 필수적이지만 CDK6는 특정 세포에서만 작용하며 CDK2가 역할을 대신할 수 있습니다.  CDK 4/6 모두의 inhibitor는 hormone receptor-positive/HER2-negative 유방암의 치료에 허가되었습니다.
