특징

면역반응의 조절은 co-stimulatory 및 inhibitory checkpoint receptor의 균형을 통해 엄격하게 제어됩니다. 따라서 억제 수용체를 차단하거나 면역자극 체크포인트 수용체를 활성화하는 치료제는 anti-PD1 요법과 같은 항종양 면역반응을 회복시키는 강력한 약제임이 입증되었습니다.
PathHunter Immune Checkpoint assay는 면역치료제의 스크리닝, 역가테스트, lot release를 위한 cell-based assay를 제공합니다.

원리

Signaling cell의 checkpoint receptor (예, PD-1, SIRPa 등) C-말단은 PK로 fusion되어 있고, SH2-domain은 EA가 tagging되어 있습니다. Ligand-presenting cell(암세포)과의 공동배양에서 ligand (예, PD-L1, PD-L2등)가 checkpoint receptor와 binding 할 경우, active한 b-gal enzyme이 형성되어 기질의 가수분해를 통해 화학발광 합니다. 만일 항체나 small molecule에 의해 PD-1과 PDL-1의 결합이 차단될 경우에는 빛이 형성되지 않습니다.
