바이오시밀러의 functional protein level을 측정하기 위한 ARRAY BRIDGE 사의 PEP Assay Kit 입니다. Protein Kinases, Protein Phosphatases, Proteinases, Oxido-Reductases 등에 대한 기능을 확인하고 해당 단백질을 정제합니다.

특 징 

 

  • 단백체로부터 수백-수천가지에 이르는 기능 단백질을 동시에 측정하고 정제하기 위한 킷트 입니다. (정제된 단백질은 Mass Spectrometry로 동정할 수 있습니다.) 
  • 단백질 기능을 유지한 채로 2D 전기영동을 진행한 후 Protein Elution Plate (PEP)에 옮겨서 Enzyme Assay 및 Protein purification을 진행합니다. 
  • Proteomics에서 functional protein level을 연구하는데 효과적입니다.
  • Protein Kinases, Protein Phosphatases, Proteinases, Oxido-Reductases 등에 대한 기능을 확인하고, 해당 단백질을 정제합니다. 
  • Small Format은 384 well kit으로, Large Format은 1536 well kit (384well plate x 4개)으로 제공됩니다.
  • 모든 Protein Elution Plate에는  6,000 dalton의 MWCO 막이 부착되어 있어 분자량이 6,000 dalton 이상인 단백질을 모을 수 있습니다.
  • 단백질이 젤에서 PEP로 이동 된 후 단백질 확산을 감소시키기 위해 특수 용액이 제공됩니다.

과 정 




  1. 샘플 준비 
  2. IEF gel 분리 및 2D 전기영동
  3. PEP plate를 준비하고 gel 상의 단백질을 PEP로 transfer
  4. 384 well plate를 준비하고 PEP plate로부터 protein recovery
  5. Enzyme assay
  6. Data reading 
  7. Enzyme activity가 있는 well이 발견되면 해당 well에 존재하는 단백질의 순도 체크 (젤 running 후 형광염색 및 CCD imaging)
  8. Mass spectrometry로 타겟 protein 확인 동정
