A. Insect 발현 시스템

 

곤충세포 발현시스템은 baculovirus에 감염된 곤충 세포에서 virus 생산 및 제조합단백질을 발현합니다. 이 발현 시스템은 단백질 용해도를 향상시키고 복잡한 PTM을 잘 수행합니다. 동시에 여러 유전자를 발현하여 이종이량체 또는 중합체를 형성하고 더 높은 수율로 세포내 단백질 및 분비 단백질을 생산할 수 있습니다.

이 시스템은 kinase, viral target, cell surface receptor, transporters, enzymes membrane protein과 같이 발현하기 어려운 단백질의 생산에 효과적입니다. 

막 단백질의 기능은 질병 상태와 관련이 많기 때문에 현재 약물 표적의 약 60% 이상을 차지합니다. 이러한 생물학적 관련성에도 불구하고, 이러한 단백질을 제작하고 정제하는 문제로 인해 제약이 많습니다. 곤충세포 발현시스템은 또한 400kDa 이상의 large insert를 발현할 수 있습니다.

B. Bacteria 발현 시스템

 

E.coli는 cytokine, enzyme, immunogen, antibody fragment 등 광범위한 재조합 단백질에 이용됩니다. 제작비용이 상대적으로 경제적이며, 유전자 조작이 쉽다는 장점을 가지며, Protein yield가 15g/L 이상으로 높고, Kg 수준의 scale up이 가능한 발현 시스템입니다.
