GPCR Functional Assays


  • GPCR 치료제의 잠재적 효능과 기능을 평가할 때 on-and-off target activity를 이해하는 것이 중요합니다. GPCR은 다양한 cascade를 통해 신호를 보낼 수 있으며, 약물은 이들 중 하나 이상을 활성화, 억제 또는 조절합니다. 
  • GPCR Functional assay는 화합물의 활동을 더 잘 이해하기 위해 여러가지의 2차 메신저 경로를 이용함으로써 약물 후보의 in vitro pharmacology profile에 대한 평가를 제공합니다.
  • Gi 및 Gs 결합 수용체에 대한 cAMP 분석, Gq 결합 수용체에  대한 IP1 및 IP3/Calcium flux 분석, receptor의 자연 경로 분석, label free  등의 다양한 분석법을 제공합니다. 
  • FLIPR®, EnVision®, CellKey™, CellLux™등의 플랫폼을 제공합니다.
  • full, partial, inverse agonist 및 antagonist의 활성을 측정합니다.
  • EC50, IC50 측정이 가능합니다.



GPCR Radioligand Binding Assays

  • Radioligand binding 분석은 GPCR과 화합물의 상호작용을 검출하기 위한 gold standard로, 분석 결과를 가장 간단하게 해석할 수 있습니다. 
  • 직접 제작한 GPCR membrane preparation을 이용하고 Cerep, Panlab에서 개발된 전문 지식을 활용하여 신뢰도 있는 결과를 제공합니다. 
  • 125가지 이상의 타겟을 제공합니다.
  • Single concentration screening 및 IC50 determination을 위한 dose-response 조건의 스크리닝이 가능합니다. 
  • 제공되는 결과 : 
    • Percent inhibition (mean of replicates)
    • Individual values as a percent of control, EC50 or IC50 value (calculated from a minimum of 5 concentration testing)
    • Ki value (calculated using Cheng-Prussoff equation, and the true Kd of the membrane preparation used for the experiment)
    • Hill coefficient (nH)
    • Plotted EC50 or IC50 curves
