특징

Anti-CD19 CAR-T Lentivirus는 anti-CD19-ScFV (VL-VH)-CD8 hinge-4-1BB-CD3ζ 구조의 2세대 혹은 3세대 CAR(Chimeric Antigen Receptor)를 발현하는 tumor-specific premade lentivirus 입니다.
CD19 positive cancer cell을 타겟팅하여 사멸시킵니다.
Amount : 200ul/vial
Titer : 1x10^8 IFU/ml
표적 CAR-T 사멸의 특이성을 검증하기 위해 음성 대조군 CAR lentivirus를 제공합니다.
1) CAR-ctr1 : Anti-CD19-scFv 인식기능이 제거되어 있는 negative control
2) CAR-ctr2 : CD3 activation domain이 제거되어 있는 negative control
사용법 : 24well plate에 0.5ml의 T cell을 seeding하고, 1M cells/ml에 50ul의 anti-CD19 CAR-T lentivirus를 넣어 48-72시간 후 transduction을 관찰합니다.

구조

anti-CD19 (clone FMC63)의 scFv 부분이 CD8 hinge, 4-1BB co-stimulatory domain과 CD3ζ signaling domain의 CAR와 퓨전되어 EF1a promoter 하에서 발현됩니다.
Enhanced EF1a promoter를 사용하였습니다.
T cell에 잘 transduction 됩니다.
GFP, RFP 시그널을 통해 transduction 여부를 쉽게 확인할 수 있습니다.
Rsv promoter하에서 Fluorescent-Antibiotic fusion dual selection marker를 이용하여 형광 혹은 항생제로 transduced cell을 sorting할 수 있습니다.

CD19 : B세포 계통에서 백혈병 및 림프종 마커로 CD19 표적 요법의 target으로 사용됨
4-1BB (CD137) : CAR 디자인시 가장 널리 연구된 co-stimulatory domain. 활성화된 T림프구, CD4, CD8 T cell에 존재하는 표면 당단백질. 항원제시세포 (대식세포 및 활성화된 B cell)에 발현된 리간드에 결합하여 T cell의 anti-tumor activity를 촉진
CD3ζ (T-cell receptor zeta) : 가장 일반적으로 많이 사용되는 CAR의 activation component. T cell과 NK cell에 의해 발현. TCR 및 CD3γ, δ, ε 사슬과 함께 TCR-CD3 복합체 형성
CD28 : T세포에서 발현되는 transmembrane protein
