특징 

 

  • EMBER500™ RNA Prestain Loading Dye는 RNA denaturation, loading, staining을 한번에 할 수 있는 all-in-one loading dye입니다.
  • EMBER500는 매우 밝은 DNA/RNA 결합 염료로 UV 및 blue LED에서 사용할 수 있습니다. 
  • EMBER500 prestaining은 EtBr prestaining 보다 훨씬 더 밝은 신호와 높은 감도를 제공합니다. 
  • 포름아미드 (formamide)가 함유되어 있어 RNA를 일반 아가로스 젤에서 전개할 수 있습니다. 
  • RNA 정제 샘플에서 gDNA를 감지할 수 있고, RNA의 무결성을 검증할 수 있습니다.
  • EMBER500™ prestaining은 저분자량 RNA나 ssRNA 분석, denaturing gel 염색에는 권장되지 않습니다. 이 경우에는 Oxazole Gold (#40094)로 post-staining 할 것을 권장합니다.

1. RNA를 로딩했을 때, EtBr에 비해 월등히 밝습니다.


 

Figure 1. Human total cellular RNA in water was mixed with EMBER500™ RNA Prestain Loading Dye or loading dye with 250 ug ethidium bromide (EtBr). Samples were heated for 10 minutes at 70°C, then run on a non-denaturing 1% agarose/1X TBE gel. From left to right, loading amounts were 200, 100, 50, or 25 ng/lane for total RNA. Lanes 1-4 used EMBER500™ RNA Prestain Loading Dye, lanes 5-8 used RNA loading dye with EtBr. The gel was imaged using a UVP GelDoc-iT® imaging system with a UV transilluminator with EtBr filter with 1.5 second exposure time.

 

 


2. RNA와 DNA 모두 염색하여, 정제된 RNA샘플에서 gDNA 오염을 감지하고, RNA 무결성을 검증할 수 있습니다.

3. 일반적인 필터를 사용하여 UV transilluminator에서 볼 수 있고, UV 노출 위험을 줄일 수 있는 LED에서도 관찰이 가능합니다.
