특징
Claudin 18.2 (CLDN18.2)는 최근 항암제 연구, 특히 위암 및 췌장암 치료 분야에서 가장 주목받고 있는 표적 단백질(타겟) 중 하나입니다. Claudin 중에서도 18.2번(CLDN18.2) 아형은 매우 독특한 발현 특징을 가지고 있습니다. 정상 조직에서는 오직 위점막의 분화된 세포에서만 제한적으로 발견되거나 세포 내에 깊숙하게 숨겨져 있지만, 위암, 췌장암, 식도암, 담도암 등이 발생하면 세포 구조가 무너지면서 숨겨져 있던 CLDN18.2가 세포 표면 밖으로 훤히 노출되므로, 정상 세포는 다치지 않게 보호하면서 암세포만 정밀 타격하기에 완벽한 조건을 갖추고 있습니다.
Tight junction은 상피세포 사이의 세포 간 공간을 통과하는 분자의 흐름을 조절하는 세포 간 장벽을 형성합니다. Claudin (클라우딘)은 이 타이트 정션의 핵심 구성 요소로, 약 20~27 kDa 크기의 막단백질이며 보통 4개의 막 관통 도메인과 2개의 세포 외 루프를 가집니다. 약 27종의 클라우딘 단백질이 조직별로 다르게 분포하며, 이들의 기능 이상은 여러 암과 연관되어 있어 진단 및 치료의 잠재적 표적이 됩니다.
- Claudin 18 isoform 2 (CLDN18.2)는 정상시에 위 점막 상피세포에서만 발현되는 단백질이지만, 선암, 췌장암, 식도암, 폐암등과 같은 특정암 발생시 과발현됩니다. 이러한 비정상적인 발현은 CLDN18.2가 종양의 침습 및 전이에 중요한 역할을 할 수 있음을 시사합니다.
- CLDN18.2는 일차 위선암과 그 전이암의 50~70%에서 발현되기 때문에, 위암 표적 치료의 유망한 후보로 간주됩니다.

Then the cells were fixed with paraformaldehyde for 15 minutes. After washing, the cells were incubated with gradient concentration of IMAB-362 (anti-CLDN18.2 antibody) for 1 hour, and incubated with goat anti-human IgG (H&L) [HRP] antibody. After incubation with TMB substrate, OD450 was measured by a microplate reader.
