Lentivirus를 이용한 Stable Cell Line 제작서비스 

• 표적 유전자를 lentivector에 subcloning하여 lentivirus particle을 생산한 후 세포주에 도입합니다. 
• 유전자 발현을 최적화하고 gene silencing을 피하기 위해 다양한 promoter를 이용합니다.
• Reporter, antibiotic selection marker, promoter등을 직접 선택하여 원하는 디자인의 vector backbone을 제작합니다.
• Inducible expression cell line의 경우, 먼저 TetR 억제인자 발현 입자로 세포를 감염시켜 TetR stable cell line을 만든다음, 타겟을 발현하는 파티클로 감염시켜 tetracyclin-inducible stable cell line을 생성합니다.
  
• GenTarget사의 독점적인 suCMV promoter는 매우 강력한 발현 효율을 가지며 타겟에 대해 높은 수준의 발현을 제공합니다. 

[제작 과정]

1. Construct a lentiviral vector containing your gene and desired selection marker
2. Generate lentivirus
3. Transduce the cell line you request
4. Select the stably transduced, high-expression cells
5. Validate integration of the gene via genomic PCR
6. Validate the highest-expressing clone by Western Blot (if applicable)
7. Deliver two cryogenic vials of stable cells along with a final report

• CRISPR/Cas9 기술을 이용하여 gene knockout이 도입된 세포주를 제작해 드립니다.
• 90여 가지의 popular transfection-suitable cell lines에 적용 가능 합니다. (예, A549, CHO-K1, HEK293, HEK293T, HT-29, MDA-MB-231, 4T1, A20, HCT116, MCF7, MDCK, U937, RPMI 8866 등)

1. Host cell line preparation, gRNA design & synthesis (3-4주)
2. Cell pool generation (1-3주)
3. Single cell cloning (2-4주)
4. Screening & Validation (2-3주)
5. Clone cryopreservation & QC reports (2-6주)
