​Tyramide signal amplification (TSA)란? 

형광 염료가 표지된 항체를 사용하는 기존의 방법과는 달리, TSA는 Tyramide의 활성화를 촉매하는 HRP(horseradish peroxidase)가 부착된 항체를 사용합니다. HRP에 의해 활성화된 Tyramid-형광 or Biotin은 근처 조직의 tyrosine 잔기에 공유 결합하여 부착됩니다. TSA를 이용하면 IF를 이용하는 것보다 밝은 형광 신호를 얻을 수 있으며, 항체의 cross-reactivity에 영향을 받지 않습니다. 

• 미량의 타겟도 감지할 수 있습니다.
• ICC, IHC 및 FISH와 호환됩니다.
• 기존의 방식의 최대 100배 수준으로 민감합니다.
• 기존의 ICC, IHC 및 FISH와 workflow가 유사합니다.
• 소량의 1차 항체를 사용합니다.
• Multiplex IHC에 효과적입니다. 

Tyramide signal amplification(TSA) system. A cell or tissue sample is labeled with primary and secondary antibody using conventional methods. The horseradish peroxidase, conjugated to the secondary antibody, catalyzes the conversion of labeled tyramide into a reactive radical. The tyramide radical then covalently binds to nearby tyrosine residues, providing high-density labeling.

CF® Dye Tyramide

실험 결과

Multiplex tyramide labeling of FFPE tissues. Cytokeratin (pan) was labeled with CF®488A-tyramide (green); Histone H1 was labeled with Cy®3-tyramide (red); ZO1 was labeled with CF®640R-tyramide (magenta).

Sequential labeling of formaldehyde-fixed HeLa cells with two Tyramide Amplification Kits. Mitochondria (green) was labeled with rabbit anti-COXIV primary antibody and Tyramide Amplification Kit with HRP goat anti-rabbit IgG and CF®488A-tyramide, followed by peroxidase quenching. Nucleoli (magenta) visualized with mouse anti-cyclin B1 primary antibody and Tyramide Amplification Kit with HRP Goat anti-mouse IgG and CF®568-tyramide.  

Staining more targets simultaneously allows detailed studies of tissue architecture. Automated spatial analysis (CytoMAP) of 12-plex CF dye staining of myeloid markers in lymph nodes (LN) reveals myeloid organization.
