GPCR Functional Assays

Pharmacology of several adrenergic compounds at the alpha1A adrenergic receptor. Four agonists and antagonists were tested in a cellular Ca2+ mobilization assay 
using CHO cells transfected with the human alph1A adrenergic receptor. The endogenous ligand epinephrine and alpha1 antagonist WB 4101 are used respectively 
as standard reference agonist and antagonist compounds for the assay and tested in each experiment with client compounds. Data are expressed as % of control epinephrine response.

gpcrBIAS 분석 서비스 컨셉 

It has been found that receptors are not simply switches that uniformly activate cell signaling pathways but rather are microprocessors able to bias activation to some signaling pathways at the expense of others (Kenakin 1995). This phenomena changes the overall quality of efficacy possessed by an agonist. Ligand bias can lead to therapeutic opportunities or potential liabilities and is illustrated with analogs of angiotensin above.


GPCR Radioligand Binding Assays

  • Radioligand binding 분석은 GPCR과 화합물의 상호작용을 검출하기 위한 gold standard로, 분석 결과를 가장 간단하게 해석할 수 있습니다.

  • Single concentration screening 및 IC50 determination을 위한 dose-response 조건의 스크리닝이 가능합니다.

    • Percent inhibition (mean of replicates)
    • Individual values as a percent of control, EC50 or IC50 value (calculated from a minimum of 5 concentration testing)
    • Ki value (calculated using Cheng-Prussoff equation, and the true Kd of the membrane preparation used for the experiment)
    • Hill coefficient (nH)
    • Plotted EC50 or IC50 curves


Pharmacology of several adrenergic compounds at the alpha1A adrenergic receptor. The alpha1 antagonists WB 4101 and prazosin, alpha1 agonist cirazoline and 
endogenous ligand epinephrine were tested in a binding assay using [3H]prazosin (0.1 nM) as a radioligand and membranes obtained from CHO cells transfected 
with the human alph1A receptor. WB 4101 is used as a standard reference compound for the assay and tested in each experiment with client compounds. 
Data are expressed as % of control specific [3H]prazosin binding.
