특징

ADCC(Antibody-dependant-cell mediated cytotoxicity, 항체 의존 세포 매개 세포 독성)는 암이나 바이러스가 감염된 타겟 세포를 용해하기 위한 면역세포의 활성화를 유도하는 세포 매개 면역 방어 메커니즘입니다.
Test compound를 target cell (BT-474 cancer cell line)에 인큐베이션 한 후, effector cell (PBMC)를 추가하여 현미경으로 live cell 이미지를 관찰하고, target cell에 대한 cytotoxicity를 형광으로 측정합니다.
검증된 Anti-HER2 Drug인 Trastuzumab를 control로 사용합니다.
다양한 표적 및 effector cell을 이용한 맞춤형 ADCC 분석을 개발해 드립니다.

Testing Information

Procedure Summary: Test compound and/or vehicle is preincubated with Calcein AM-stained Target cells (BT-474) at 2.5 x 10^5 cell/ml (1 x 10^4 cells/well) for 30 minutes, followed by adding effector cells (PBMCs*) with an E:T ratio of 20:1 for 20 hours co-culture. Live cell images are acquired by ImageXpress Micro Confocal Image system and cytotoxicity of target cells is quantified by fluorescence intensity. Decrease of fluorescence intensity by 20 percent or more (≥ 20%) relative to 1 μg/ml hIgG1 isotype control indicates possibility of ADCC ability.
(*In consideration of donor-dependent variations, three donors are suggested in every test to ensure optimized signals are obtained.)
Incubation: 1200 min at 37°C
Control Inhibitor: Trastuzumab
Criteria For Significance: ≥ 20% decrease in fluorescence intensity relative to hIgG1 isotype control
Test Concentration / Dose: Compounds are screened at 10, 3, 1, 0.3, 0.1, 0.03 and 0.01 μM.
Test Sample Requirements: Minimum for 1) Screen: 25 μl of 10 mM stock -OR- 1 mg (pre-weighed). 2) Dose Response: 60 μl of 10 mM stock -OR- 1 mg (pre-weighed).
