특징

 

  • 19개의 서로 다른 조직 유형, 300개 이상의 2D 암세포주와 100개 이상의 3D 종양 spheroid 모델에 대한 항암제 후보 물질 약물 반응 효과를 스크리닝 합니다.  

  • mRNA 과발현, 증폭, 삭제, 코딩 및 비코딩 돌연변이, 기능 돌연변이 획득 또는 손실을 포함하여 250,000개 이상의 유전적 이상에 대한 데이터 기반 통찰력을 얻을 수 있습니다. 

  • 생성된 데이터는 환자의 치료에 대한 반응을 예측하거나 잠재적인 내성 기전에 대한 게놈 특이적 바이오마커 및 유전자형을 식별하여 암세포에 가장 효과적인 약물을 도출할 수 있습니다.

  • Singleplex 
    • Cell proliferation - including IC50, EC50, and GI50, integrated area values and intra-assay doubling time.
  • Multiplex
    • Cell proliferation - including IC50, EC50, and GI50, integrated area values and intra-assay doubling time.
    • Apoptosis - concentration required for significant caspase-3 induction (5-fold and 6SD over vehicle) and maximum fold induction (Emax).
    • Cell cycle - concentration required for significant increase or decrease (2-fold relative to vehicle) in histone H3 phosphorylation.
  • Univariate Genomic Analysis
    • Identify and prioritize predictive genomic biomarkers from drug response data by analysis of hundreds of thousands of genomic features.  Genomic features that are analyzed include mRNA overexpression, amplifications, deletions, coding, and non-coding mutations and gain/loss of function lesion categories.  Analysis in the context of cancer subtypes and tissue classification.

A database of genomic features evaluated for correlation to PD0325901 responsiveness, as measured by cell count IC50. Use of orthogonal Student’s t-test (x axis) and Fisher’s exact test (y axis) address mathematical bias, allowing identification of biologically relevant features significant by both. Genomic features satisfying both significance thresholds, and with q-values (FDR) <0.1, are colored dark green and annotated.
