Viability PCR (v-PCR)이란?

• Viability PCR은 살아있는 미생물의 신속한 검출을 위한 기술로, 시간이 많이 걸리는 배양 방법과 달리 빠르고 민감합니다.
• 일반적인 qPCR 방법이 살아있는 세포와 죽은 세포를 구별하지 않는 반면, Viability PCR은 살아있는 미생물만 증폭하므로, 감도 및 특이성이 높습니다. 
• 박테리아 뿐 아니라 효모, 바이러스, 진핵생물, archaea 및 다른 유기체에도 적용할 수 있습니다. 
• PMAxx와 PMA는 DNA에 대한 높은 친화력을 갖는 photoreactive dye로, dsDNA에 삽입된 후 가시광선에 노출되면 공유결합을 형성합니다. 살아있는 세포에서는 세포막을 투과하지 못하므로, 세포막이 손상된 죽은 박테리아의 DNA에만 결합하여 DNA 변형을 유발함으로써 높은 Ct와 증폭 지연을 유도하여, live cell과 구분됩니다. 
  

Viability PCR dyes like PMAxx™ or PMA are membrane-impermeant, which makes them dead cell specific. Once inside of a dead cell, they bind to DNA. Exposure to intense visible light renders the dyes reactive, and causes them to covalently attach to the DNA. This DNA modification prevents amplification in subsequent PCR reactions. 

The basic v-PCR workflow involves the following 5 steps: 1) dye addition   2) incubation   3) light exposure   4) DNA extraction   5) qPCR 

PMA (#40019) & PMAxx™ (#40069)

• PMA (propidium monoazide)는 dsDNA에 high affinity binding 하는 photo-reactive dye 입니다.
• PMA dye는 dead cell의 membrane만 통과하여 dsDNA에 binding되고 visible light에 의해 DNA를 변형시킵니다. 이렇게 modified 된 dsDNA는 qPCR 진행시 증폭되지 않습니다. 즉, 살아있는 bacteria만 PCR이 진행됩니다.
• PMAxx는 PMA에 비해 superior activity를 갖는 new dye로, dead cell의 PCR을 막아 위양성 위험을 최소화 합니다.

Live or dead bacteria treated with either PMA or PMAxx™ prior to qPCR. Dead bacteria treated with either PMA or PMAxx™ show delays in amplification compared to either live cells, or untreated dead cells. Bacteria treated with PMAxx™ show a larger delay than those treated with PMA. Therefore PMAxx™ is better able to differentiate live vs dead bacteria by viability PCR.

PMA Enhancer (#31038)  

• 살아있는 그람 음성균과 죽은 그람 음성균의 구별을 위해 이용됩니다. 
• Enhancer를 처리한 시료에서는 죽은세포의 신호가 감소합니다. 
  
• PMA, PMAxx에 호환됩니다.  

E. coli were killed with mild heat treatment (56°C for 3 hrs) and treated with PMAxx™ or PMAxx™ + Enhancer, followed by light exposure using PMA-Lite™, DNA purification, and qPCR with Fast EvaGreen® qPCR Master Mix. dCt values were calculated by subtracting the Ct without dye from the Ct with dye. Only dead cells treated with PMAxx™ + Enhancer showed a large dCt, indicating that the dye successfully inhibited PCR of dead cell DNA.
