HMGB1이란?

• HMGB1 (High Mobility Group Box 1)는 핵내에서는 gene transcription, gene regulation, DNA repair에 관여하는 DNA-Binding protein으로 기능하고, cytosol에서는 Beclin-1과 상호작용하여 autophagy를 유도합니다. 
• HMGB1이 괴사등으로 세포밖으로 유리되면 chemokine이나 DAMP(Damage-associated molecular pattern)로서 염증 반응을 유도하여 면역세포를 활성화 합니다. 
• HMGB1은 TNF, IL-1 및 IL-6을 포함한 전 염증성 사이토카인의 방출을 촉진합니다.
• 혈청 내 HMGB1 상승은 COVID-19 환자의 높은 사망률 및 폐 사이토 카인 폭풍과 관련이 있다고 보고되었습니다. 이는 HMGB1이 중증 COVID-19에 대한 바이오 마커임을 시사합니다.  
• HMGB1은 종양미세환경 (TME)에서는 종양성장, 혈관신생, 면역억제, 침습, 전이를 촉진하여 Pro-tumoral protein으로서 작용하면서도 화학방사선요법 (chemotherapy)환경에서는 면역원성 세포사멸을 매개함으로써 항종양 면역을 향상시킵니다. 따라서 HMGB1은 암화학방사선요법 및 면역요법에 대한 잠재적인 치료 전략을 제공합니다.
