특징


  • 정확한 유세포 분석 데이터를 수집하기 위한 calibration, compenation, cell counting 품질관리 용 control bead를 제공합니다. 
  • Quality control bead를 사용함으로서 기기의 변동성을 최소화하고 정밀도를 높일 수 있으며,  장비 간 데이터나 사용자 간 데이터의 차이를 보정하여 보다 신뢰할 수 있는 결과를 제공합니다. 


장비 보정을 위한 Rainbow Calibration Particles

  • 유세포 분석기(flow cytometer)의 일관성 유지 및 정확한 설정, 민감도, 선형성(linearity) 확인을 위해 사용되는 control bead 입니다.
  • 입자 크기와 형광 강도가 정해져 있는 calibration particle을 사용하여 유세포 분석기를 calibration 합니다. 
  • 형광 강도를 임의 단위에서 MESF (Molecules of Equivalent Soluble Fluorochrome) 단위로 정규화하여, 일일 변동이나 장비 간의 차이를 보정할 수 있도록 합니다.
  • 자외선 (UV) 및 적외선 (IR)에 이르는 다양한 형광 강도를 가진 유사한 크기의 입자들로 구성되어 있습니다.

간섭현상 보정을 위한 Compensation Beads

  • 여러 형광 사용 시 형광 신호끼리의 간섭 현상(spillover)을 보정하는 데 사용되는 control bead 입니다.
  • Gating과 전압 설정을 안정적으로 수행할 수 있으며, 샘플 사용량을 줄여 세포를 절약할 수 있습니다.
  • Compensation bead로 매개변수를 설정하면 검출기가 적절한 형광색소의 형광 방출만 측정하여 분석의 신뢰도를 높입니다. 
  • 형광 보정을 위해 anti-mouse IgG가 붙은 positive bead 및 음성대조군으로 사용하는 negative bead로 제공됩니다.

Anti-mouse IgG 가 코팅된 positive control bead는 형광항체와 결합하여 일정한 형광을 나타내고, negative control bead는 형광항체와 결합하지 않습니다.

세포수 계산을 위한 Rainbow Cell Counting Beads

  • Rainbow Cell Counting Beads는 유세포 분석기를 이용해 세포 수를 정확하게 계산할 수 있도록 도와주는 bead입니다.
  • bead의 수가 정확히 정해진 상태로 제공되어, 세포 시료와 정확한 비율로 섞은 후 유세포 분석기로 같이 측정하면, bead 및 세포가 몇개 검출되었는지 확인하고, 그 비율을 계산하여 시료 속 세포 수나 농도를 추정할 수 있습니다. 
  • FITC, PE, PE-TR, PE-Cy5 및 APC를 포함한 여러 형광 채널에서 사용할 수 있습니다.

NK 세포의 K562 세포 살해 능력(NK cell killing activity)을 정량적으로 평가하기 위한 실험으로, 14일간 배양된 NK 세포와 K562 세포를 여러 비율(K562:NK)로 혼합하여 세포 살해 정도를 분석하였습니다. K562 세포는 Mito-Mark Green으로, NK 세포는 Janelia Fluor® 646으로 각각 염색하였습니다. Rainbow Cell Counting Beads [NBP3-00495]를 함께 사용하여, 유세포 분석기(FACS)로 얻은 데이터를 정규화하고 정확한 세포 수 및 살해율을 계산하였습니다. 그 결과, K562 세포만 단독으로 존재할 때는 세포 살해가 관찰되지 않았으며, NK 세포를 5:1 비율로 추가한 경우에는 Mito-Mark Green 신호가 감소하며 세포 살해가 명확히 확인되었습니다. 이는 NK 세포가 K562 세포를 효과적으로 사멸 시켰다는 증거로 해석됩니다.
