원리

이 분석법은 태그(anti-tag) 항체로 코팅된 마이크로플레이트를 사용합니다. 표준물질(standards), 샘플(samples), 대조군(controls)을 플레이트에 넣고, 이후 항체 칵테일(antibody cocktail)을 추가합니다. 1시간 동안 반응/incubation한 후, 플레이트를 세척하고 기질(substrate)을 추가하여 20분간 반응시킵니다. 그 다음 정지 용액(stop solution)을 넣고, 일반적인 마이크로플레이트 리더기를 사용하여 결과를 판독합니다.

Linearity of Dilution
희석 선형성(Linearity of dilution)은 R&D Systems®의 Quantikine® ELISA에서 매우 중요한 특징이며, QuicKit™ ELISA도 예외는 아닙니다. 희석 선형성은 매트릭스 효과(matrix effect)를 평가하고, QuicKit™ ELISA가 샘플 값을 정확하게 측정하는지 확인하는 데 중요합니다. 만약 희석 선형성이 좋지 않거나, 100%에서 크게 벗어난 결과가 나온다면, 이는 매트릭스 효과로 인해 분석 대상(analyte)의 검출이 인위적으로 증폭되거나 방해를 받았을 가능성을 나타냅니다.



Spike and Recovery
스파이크 및 회수 실험(spike and recovery experiments)에서는 자연 샘플(natural samples)에 재조합된 분석 대상 물질(recombinant target analyte)을 첨가하여, 샘플 매트릭스(matrix)로 인한 간섭(interference)이 있는지를 확인합니다. 스파이크 및 회수 실험은 측정값의 정확도(accuracy)를 평가하기 위한 중요한 도구입니다.
