특징 

 

  • 생물학적 제제의 생산 과정에서 사용된 세포의 단백질(Host cell protein)을 측정하는 데 사용되는 ELISA Kit 입니다. 
  • CHO, E. coli, HEK293, Pichia Yeast로부터 생산된 단백질이나 항체등, 바이오 의약품 개발 및 제조에서 품질 관리시, HCP의 농도가 안전한 범위에 있는지 모니터링 하기위해 사용됩니다. 
  • Cell lysate, cell culture supernatants, 단백질 정제과정, 최종산물 등에서 HCP 잔류물질을 확인합니다. 
  • ISO13485에서 생산됩니다. 


원리 

Microplate는 capture 항체로 미리 코팅되어 있습니다. 측정할 항원과 HRP 결합 탐지 항체가 순차적으로 Microplate의 well에 추가되어 capture 항체-항원-탐지 항체 복합체를 형성합니다. 세척 후, 반응에 관여하지 않은 결합체들은 제거되고, TMB 기질 용액이 웰에 추가되어 인큐베이션이 진행됩니다. TMB 기질 용액은 HRP의 산화 후 파란색으로 변하며, 마지막으로 정지 용액을 추가하면 즉시 노란색으로 변합니다. TMB 기질의 색은 초기 단계에서 결합된 항원과 양의 상관관계를 가집니다. Microplate 리더기를 사용해 450nm에서 흡광도(OD 값)를 측정하고, 이를 기준으로 표준 곡선과 해당 농도를 작성합니다. 그런 다음 샘플의 OD 값을 표준 곡선 방정식에 입력하여 샘플 내 목표 단백질 농도를 계산합니다.
