U-CyTech사의 T cell ELISpot Kit은 단한개의 세포가 분비한 cytokine도 검출해 낼 수 있는 매우 민감한 면역 검사법으로, cytokine을 생성하는 세포를 확인하고 측정하는데 이용됩니다.

특징

 

  • Lymphoid tissue, CNS tissue, bone marrow, PBMC 등의 single cell suspension에서 cytokine을 분비하는 T cell을 확인합니다.
  • Cytokine이 cell에서 분비 되는 순간 antibody에 바로 binding 하므로 적은양의 cytokine도 detection 됩니다.
  • 사이즈 : 2-plate, 5-plate kit
  • Plate  : transparent polystyrene-bottomed, PVDF membrane-bottomed (PVDF) plate 방식으로 제공
  • coloring agents : silver (black spot), enzymatic (red spot) 방식으로 제공
  • 10-plate를 제작할 수 있는 antibody pair (coating + detection antibody)도 별도로 구매하실 수 있습니다.

실험 과정

T Cell ELISPOT Kit 원리

1. coating antibody가 코팅된 well plate에 세포를 첨가한다.
2. 이 세포가 cytokine을 분비하도록 (항원을 넣어) 감작시키고 배양한다.
     (배양하는 동안 세포가 분비한 cytokine은 capture antibody에 binding 된다.)
3. 세포를 lysis 시켜서 제거하고 biotinylated detection antibody를 넣는다.
4. enzyme-labelled streptavidin 혹은 anti-biotin antibody를 넣는다.
5. substrate를 넣고 spot이 형성되는 것을 관찰한다.
    (cytokine을 분비한 세포에만 발색 spot이 보이므로, 발색된 cell을 카운팅하면 어느정도의 세포가 항원 자극에 반응했는지를 측정할 수 있다.)
