BCECF는 pKa (6.97)가 생리적인 pH와 가깝고, 세포질의 pH 변화를 민감하게 detection 할 수 있기 때문에 가장 널리 사용되는 형광 pH indicator입니다.

  • BCECF acid (Full chemical name: 2',7'-bis-(carboxyethyl)-5-(and-6)-carboxyfluorescein) (purity > 95% by HPLC)
    • Catalog Number: 51010
    • BCECF (full chemical name: 2¢ ,7¢ -bis-(carboxyethyl)-5-(and-6)-carboxyfluorescein) is the most widely used fluorescent pH indicator. Because its pKa (6.97) is close to physiological pH, BCECF can detect cytosolic pH change with high sensitivity. At low pH, the dye is weakly fluorescent but becomes more fluorescent with increasing pH. The excitation spectrum of the dye undergoes a slight shift during pH change, while the wavelength of the emission maximum remains unchanged. The pH is determined ratiometrically by the relative fluorescent intensities at 535 nm when the dye is excited at 439 nm and 505 nm respectively. BCECF acid is membrane-impermeant and therefore has to be loaded into cells via microinjection or scrape loading (See 51011, BCECF/AM).

    • This dye can be used as a low pH indicator similar to 51015, and is also useful as a general green fluorescent label. Carboxy-2’,7’-dichlorofluorescein is significantly more photo-stable than carboxyfluorescein or FITC.

      Yellow solid soluble in DMSO, DMF or basic water (pH >7). Store at room temperature and protect from light.

  • 5-(&6)-carboxy-2',7'-dichlorofluorescein diacetate (purity > 95% by HPLC)

    • Membrane-permeant and thus can be loaded into cells via incubation. The diacetate is readily hydrolyzed to 5-(and-6)-carboxy-2¢ ,7¢ -dichlorofluorescein (See 51015) by intracellular esterases.

    • Carboxyfluorescein has a pKa of 6.5 and can be used as a pH indicator. The excitation spectrum of the dye responds to pH similarly to that of BCECF (see 51010). Carboxyfluorescein can also be used as a tracer dye. The dye is membrane-impermeant and can be loaded into cells by microinjection or scrape loading.
