    • Coelenterazine (native form) is a luminescent enzyme substrate for apoaequorin and Renilla luciferase. The aequorin complex has been used as an excellent calcium indicator that can measure a broad concentration range of calcium from ~0.1µM to >100 µM.1-4 Renilla luciferase, encoded by the luc gene, has been widely used as a repoter protein. Renilla luciferase /coelenterazine has also been used as the bioluminescence donor in bioluminescence resonance energy transfer(BRET)5 to study protein-protein interactions. Other uses of coelenterazine include chemiluminescent detection of superoxide anion and peroxynitrite in cells or tissues.6-9 Biotium offers a large selection of coelenterazine analogs. Our coelenterazine e is a superior alternative to coelenterazine (native) as a substrate for Renilla luciferase. Yellow solid soluble in MeOH or EtOH.

    • Coelenterazine e is a synthetic derivative of coelenterazine, and is a superior substrate for Renilla luciferase. Coelenterazine e generates nearly 40% more light than native coelenterazine does while the initial light intensity of the former is more than 7 times higher than that of the latter.1 The emission spectrum of coelenterazine e/Renilla luciferase has two peaks centered at 418 and 475 nm, respectively (See Figure 1).

      Coelenterazine e is also a good substrate for apoaequorin and, compared with coelenterazine (native), the e-aequorin complex formed from coelenterazine e has a faster luminescence rise time and shows two emission peaks at 405 and 465 nm respectively, with the ratio of the peak heights dependent on calcium concentration in the range of pCa 5-7.2 The dual emission makes coelenterazine e/apoaequorin possible to be used for calcium detection using the ratio of the two emission intensities.     • Luciferin is a natural compound isolated from fireflies and is a substrate for the enzyme luciferase. Chemiluminescence at 560 nm is generated when luciferin is oxidized by an ATP-dependent process, catalyzed by luciferase. Luciferase is encoded by the luc gene, which is widely used as a reporter gene in a variety of cells. Because of the intrinsic low background of chemiluminescence technique, detection of the luc gene expression can be made at very low level. In addition, luciferin/luciferase has been used to measure 10-15 molar quantity of ATP.
